If something has no males at all, then there's a decent chance it's meant to be read by them. Unless it's in Yuri Hime/Yuri Shimai.

Azumanga Daioh belongs to the class of manga/anime known as 'healing' in Japan. Believe it or not, Azumanga Daioh, Hidamari Sketch, Kamichu!, and Yokohama Kaidashi Kikou are all hardcore seinen.
Old guys falling asleep on the train after working eight million hours like to read something light, happy, and mellow. 20th Century Boys isn't for everyone, or for every time.
If there's a reliable quality to Seinen it's that it usually has a little less manic slapstic comedy and fanservice than shounen does. Well, that, and the lack of helper hiragana beside the text. Other than that, pretty much anything goes.
That said, seinen often has more complex characters and plots, and so those of us who have read our fair share of shounen action manga tend to prefer the serious/heavy seinen stuff.

The only "criteria" it needs to be labeled Shounen or Seinen is the magazine it's serialized in. (Hello, Azumanga Daioh & Yotsuba&!)
Ta-da! Voila! A Shounen! Regardless of what you may think or want; Fact is, it's a shounen.
There are differences between Western standards and Eastern standards for viewer's. Eastern is more lenient whereas Western is more strict.
Or to put it another way, seinen and shonen (and shojo and josei) are statements about where it's serialized, and not about its content -- and you can get in trouble by inferring the latter from the former.

But included in that statement is the target audience.
Shounen magazines have furigana, to make it easier to read for the kids. Whereas Seinen magazines are purely kanji.
Bitter Virgin is kinda funny, there are comedy elements although its really dark comedy and most of its pretty serious drama...
